Spero Therapeutics Inc ($SPRO) recently announced a significant $105 million non-dilutive, non-recourse royalty financing deal with Healthcare Royalty affiliates, majority owned by KKR. This financing, backed by future milestone and royalty payments from Utebzi — the first oral carbapenem antibiotic approved by the FDA for complicated urinary tract infections — extends Spero’s cash runway into the second half of 2029. The company will repay the financing through quarterly payments sourced exclusively from royalties paid by GSK, which holds exclusive rights to develop and commercialize Utebzi globally, excluding select Asian markets.

Alongside this financing, Spero secured exclusive worldwide rights (outside Greater China) to SP001, a third-generation anti-CD40L monoclonal antibody licensed from Innovent Biologics, targeting immune-mediated diseases such as IgG4-related disease. The company plans to initiate a Phase II trial for SP001 in mid-2027, aiming to enroll patients in an open-label study with two dosing arms. To strengthen clinical leadership, Spero appointed Debra Jeske Zack as Chief Medical Officer, bringing over 25 years of experience in immune disease therapies. These strategic moves position Spero to advance its immunology pipeline while maintaining financial stability well into 2029.
